  3. Sooooo, the action of eluting the antibody off the red cell can change the antibody structure, i recently saw this with an Anti Fya. I dont know the exact mechanism but imagine it has to do with the very low pH causing a change in the epitope.
  4. It was Polyclonal material, I didnt detect the anti C or (G) on incoming inspection but did elute it off R2R2 Cells along with an Anti D. These days i stick to monoclonals.
  5. Yes Anti G is very likely, BUT I once manufactured Anti D coated cells using R2R2 cells and was able to elute off a weak anti C along with an anti D, it was an odd antibody as it only reacted in Gel... i still believe it was an example of M-O.
  6. Did you detect another (different) antibody in the eluate? eg. like an anti D ? If so you may want to consider Matuhasi-Ogata phenomenon....
  7. Heterozygous versus Homozygous expression for the antigen is key especially in the MNS system. You should know what the status is of the cell you are using so that you can compare apples to apples.
  8. Be aware that most Anti AB these days is a blend of anti A and Anti B monoclonal and is not strictly an anti AB.
  9. Dear BBSLM, I am happy to walk you through some hints and tips in using the Hemo bio kit with gel cards, we use it all the time with Grifols and Ortho Gel cards here in our testing lab. The Hemo kit is very similar to the Immucor /Gamma kit as it is an acid elution method.   10. You have to spin the final eluate very hard and for some time, try 5000 RPM for 5 minutes in a benchtop. We have validated our kit in Gel card systems. Other manufacturers kits may or may not be validated in a gel card. I suggest you read the IFU carefully and or call the manufacturer technical support.

  17. Sounds like poor advice from the IRL, I think what they meant to say was that the CD38 antigen is weakly expressed on Cord cells. The Kell system is completely removed by DTT treatment so you should give K neg blood as you have no way to exclude anti K, BTW that also means JSa, Jsb, Cellano etc. also Lu null cells do not express CD38 either.

  19. Unfortunately the answer is, it depends... If the anti CD47 is a human antibody, then it will cause a positive DAT as CD47 is expressed on most cells in the body including erythrocytes. If the anti CD47 is mouse or goat then it will not as this antibody will bind to CD47 but will not cause the same interference we see with Daratumamab. I believe recent therapies involve blocking some of the CD47 receptors.

  21. Be careful as the diluents used to suspend the cells will be different. For gel card assays you need a Low Ionic strength diluent. Diluents often contain potentiators and will have been validated for use in the appropriate card system. I am not saying they won't work and they probably will however you should perform validation to ensure you don't miss a significant antibody.

  24. CAP has a program called ABT which is antibody titre and includes IgM and IgG anti A as well as Anti D titres. if you download CAP's catalog you can see the program. Additinally API has a program that does the same thing www.api-pt.com For a method there is a method in the AABB technical manual as well as the uniform method which Dr. Auberchon authored. I believe there is a thread on pathlabtalk talking about the CAP ABT proficiency test.
