Positive manual screen

Any ideas? We ran a sample on our Gelsation the screen came out negative but there was a little (and I mean little, could of called it negative) something in one of the wells. We decided to confirm it by running the screen manually, the manual screen came out positive (2+) in one of the wells. Running an IAT and enzyme panel on the gelstation both came out negative.

Why did the manual method give a positive reaction when the automated was negative, we use the same reagents for manual and automated? And we repeated it.

When you did the test on the Gel Station, had you allowed the cells to come up to room temperature before using them? Were the cells properly mixed? Were the cells overmixed and therefore frothy? Was it with the same bottle of cells, rather than just the same lot - I presume it WAS the same lot of cells and cards? Was the plasma of the sample 'frothy' when you put it on the gel station? (You may have pipetted air bubbles) Is the QC on your gel station working?
