    Thanks very much for your reply. I am actually looking at the hardware required. ie, the equipments for doing the HLA. Could you please help me with the details used at your institute.

    There is not a single system for HLA matching.

    For transplants, there usually needs to be at least a 4 of 6 match. The DR, DQ, A, B, C, and DP regions need to be tested. DR, DQ are Class II which are mostly what I deal with so that is where most of my knowledge lies.

    For DR, high-resolution testing is needed, meaning that when all the analysis is complete, allele-level results will be obtained. First the general testing is done, which will identify the major probes that the specimen has (DRB1*01, DRB1*07, etc). Depending on the major probes involved, further subtype processing will need to happen, and possibly even secondary testing.

    Once those results have been obtained, usually by sequence-specific oligonucleotide probing, the sample might be at allele-level by this stage. Otherwise, no further processing on the SSOP side can occur and the specimen will need to be reflexed to either SBT (sequence-based typing) or SSP (sequence specific priming). Between these results and the SSOP results, allele-level results have usually been obtained and a match/no match can be determined.

    Terumo TSCD is the best in the field. I think you do not have to look for anything else. in fact there are many brands like Genesis, Hemonetics, but nothing compares to the performance and stability of TSCD.

    have you every tried to adjust the RCF of your ref centrifuge? or you just stick to speed of 850g?...our center the first spin for PRP is at spin of 1600 for 10 minutes of spin. there are instances we also experience the same presence of red cells in the plasma what we do is try to allow the red cells to settle for 30 minutes before we spin.

    I am from Kerala, India. My problem is as follows.

    Some of the bags in my centrifuge does not give me a RBC free PRP during the first spin.

    Other details. - Centrifugation - RCF=850g, 12 minutes, temp:22 DegreeC.

    Bag - Terumo Penpol Triple Bag 450ml with SAGM.

    we have been getting clear PRP for last six months, suddenly it has started giving trouble. But surprisingly only few of the bags give the problem, Not all the bags in the centrifuge ( Max.capacity 6 Bags). We ahve checked the rpm of the centrifuge and it is showing correct values as per the tachometer.

    Any suggestions from members - could it be a equipment problem?:(
