Anita SA
Can somebody give me the correct method of ABO antisera titration? Specially for anti-D IgG/IgM blend and anti-AB
Lcsmrz
Not sure what you're asking. Antisera is usually not titrated, unless you're determining the correct dilution for automated instruments. The analyzer manufacturer usually has a recommended procedure.
GilTphoto
The only reason I know of to titer ABO antisera or anti-D antisera, is for serological calibration of centrifuges. They are titered until you find a 1+ reaction, and that is the titer you use to do the calibration. These titers should be done with 6% albumin (made from a 1:4 dilution of 22% Albumin and saline). Monoclonal Anti-A and Anti-B are very strong, and you may need a titer of 1000 before reaching a 1+ reaction. Newer AABB technical manuals suggest using plasma from an A or B patient instead of antisera. The titer will not be as high.
