QC testing using Nageotte chamber

We use a product by R&D Systems (www.rndheme.com) called Leukoreduced Combined Controls.

R&D LeukoReduced RBC/PLT Control

LeukoReduced RBC/PLT control is a bi-level control product designed to monitor flow cytometer and Nageotte Chamber methods for quantification of residual leukocytes in LeukoReduced red blood cell and platelet products.

  • Packaged in 3mL pierceable ***** cap tubes

1- to use nageotte chamber the sample should be stained with turk's solution to get best results.

2- after staining, pippetting 100 microlitter into the chamber and leave it for 15 mints.

3- under the light microscope count just the first 5 rows..

4- use this equation to calculate the residual WBC's:

(WBC's count / 5) x 1000 x blood unit volume ml

WBC's count = from the chamber

5 = # of counted rows

1000 = volume of mixture(100micro of sample+900 micro of turk's solution)

blood unit volume= (weight of the blood unit - weight of empty blood bag) / 1.1 (specific gravity of RBC)

We are a small blood center (about 85K draw a year) and were just "dinged" by AABB for not performing QC on our manual nageotte chamber testing. So I guess THEY think it is necessary. We have to have most of our counts within a couple of hours for product release, so send-outs are not an option. We do not do enough of them daily to justify a flow either. I guess we will start with dmorissbb's reagent and see from there.
