Hi for all

I have a problem in the typing result of Fya,Fyb antigen by using Diamed gel Techniec. I trayred to solve but still !!

Any one have any solution for me, please?

Please could you give a little more detail as to the problems you are experiencing please?

Some times the cards (Diamed) gave us false positive or false negative results while typing for fya and fyb antigens.We tryed to solve the problem by increasing the time of incubation for 5 min more, increase the concentration of RBC, and centrifuge the cards before using.Is it right to change the proceduer like we did?, Do you have other ways to solve this problem?

Assuming that your results are genuinely false, but that the results with other grouping reagents are true, it sounds to me like a reagent problem, rather than a problem with the DiaMed gel technology itself.

Have you tried other sources of anti-Fya and anti-Fyb and tried to validate them by this technology?

The other thing that I wonder is, how old are the cells when you use them? The Duffy antigens can become quite weakened upon any lengthy storage.

Whatever you do though, DO NOT alter your incubation times or the concentration of your cells. If you do not operate according to the producer's instructions, YOU take full responsibility, and if anything goes wrong, YOU take the blame (in Law). The other thing is that changing the red cell concentration will change the equilibrium constant of the reaction, and that is one sure way to get false positives and false negatives!!!!!
