Has anyone extended their outdate beyond 24 hours of diluted cells? Also does anyone use the straight 3% in gel? Trying to rule out some suggestions made here.

  • :)Centrifuge 4 drops of 0.8% for 30 seconds. Remove supernant. Respend with 1 drop of saline. We use this frequently as needed.

I guess you could do both of those if you validated that they worked.  For the diluted cells it would depend on what the manufacturer of your diluent says.  For using 3% (12.5uL) - that would negate the lo-ionic medium that is gel (wouldn't recommend it).

Definitely wouldn't recommend the 3% cell concentration for gel. That would give you the same problems as using a too heavy cell suspension in tube testing (or gel testing) - potentially false negative results. Plus what David says about messing with the lo-ionic medium would apply.

 

When we were phasing out gel we diluted 3% antibody screening cells to 0.8% daily for about 18 months. It was part of morning QC/maintenance to make a batch that got us through the next 24 hours. It was a bit of a pain but we only made one set so it didn't really take us that long. I was glad to say goodbye to that little task.

Thank you I have a cowrker that nroought this up to our lab manager I am now having to validate it. I am skeptical but am having to do it. Hemobioscience Gel diluent allows 28 day out date! Asking for samples so I can validate it now. Hoping between the two my path with go for the one that is legit.

Hi,

I am just following up on conversion of 0.8% to 3% red cell suspensions.

Alot of agreement with Mary.

centrifuge 4 drops of 0.8% for 30 seconds, take off supernatant and resuspend with 1 drop of saline.

Could I have confirmation of centrifuge setting to use please?

I think we would all use a serologic centrifuge for this at the usual RPMs for long enough to get all the cells to the bottom, which 30 seconds is probably adequate.
