I am planning to validate selected antigen typing using gel methodology. If you would be willing to share your validation and/or procedures I would greatly appreciate having them as a resource during my validation. Any tips or info greatly appreciated!

I attempted to validate all my ag typing in gel. I was unsuccessful with Lea and Ortho's anti-M. I ran negative, heterozygous and homozygous cells with each reagent. For the non-AHG reacting abs I used buffered gel. for the AHG reactors, IgG gel. 50uL of 0.8% red cells, 25 uL of reagent antisera. Based on the pkg insert, incubation was at rt or 37 for 15 min and then 10 min spins.
