We only take out the cells when required. The package insert states the complement reactivity deteriorates during the life expectancy of the product.

We noticed that when using the Immucor complement cells with the Ortho anti-C3d, the results were non-existent even after 5-10 minutes incubation at room temperature. At first I thought the problem was with the complement cells, but after ordering Immucor anti-C3d, we noticed the reactions were stronger. After evaluating the Ortho and Immucor anti-C3d we decided to use the Immucor product. I also noticed that the Ortho product was anti-C3d, while the Immucor antisera was a combination of anti-C3d,C3b. Ortho offers an anti-C3d,C3b product, but I never evaluated this product. I figured; why not use Immucor antisera and controlling it with Immucor C3d control

New ProVue QC provided by Ortho
We've started using the AlbaQC vials 1 & 3. We feel these two vials cover proper range of results. It is unfortunate that we cannot alternate these two vials with vials 2 & 4 on alternate days. This is due to the fact we have the ProVue software determining if the QC has passed or not. Uding the QC on alternate days would entail changing the ProVue QC settings every day.

Policy for OBs
We perform ABo/Rh & antibody screen on all Rh negative mothers. Rh positive mothers are tested if we have no previous testing done during the recent pregnancy or if they have an antibody. Cord testing is performed on infants born to any Rh negative mothers and when Rh positive mothers have an antibody.

New ProVue QC provided by Ortho
We are still waiting in Canada for the new ProVue QC provided by Ortho pending our own FDA approval. How has been your experience in the US? Also, what do you do to QC the MTS IgG/C3d cards on the ProVue? We are looking at preparing concentrated O positive cells sensitized with anti-D Odif :cool:

weak anti-D reactions with QC
We’ve recently noticed that our two different lot numbers of Bioclone anti-D have been giving us a 2+ reaction with the Ortho Confidence System when we spin immediately after we add the positive QC cells with the antisera. The reaction strength is stronger if we let the cell/anti-D mixture aside for 30 - 60 seconds and then spun. I was told by Ortho that the D expression on some of their QC cells may be weaker some times thus the 2+ reaction. The QC package insert only states that a positive reaction is expected, but it doesn’t state what strength to expect. Has anyone else seen this?
