I´ll like to ask u all, about techniques that u can share with me for improve or increase reactivity of anti Fy (a). !

We miss or have weak reactions with several Fya samples each month in Gel. We can USUALLY pick these up using the two edged sword....PEG

Have you tried any of the solid phase assays from ImmucorGamma?

We use Capture R (solid phase) to mass screen our donor pools. However, we do our actual ID in Gel or tubes.

PEG is a good option to increase the possibility of picking up an Fya, but using PEG with a warm auto will possibly cause you to miss it as well as any other underlying alloantibodies.

Have you considered looking into the Solid Screen "LISS in a microwell" methodology used on the TANGO system? It has a high degree of specificity without sacrificing sensitivity. The vendor is Biotest Diagnostics based in New Jersey. Their instrument and reagents were FDA approved in 2006. They also have afull line of traditional blood bank reagents FDA approved in the fall of last year.

Whatever method you use, make sure that your cells are fresh, as the Fy(a) and Fy( antigens decrease in strength with storage, and, if you possibly can (not easy, I know) ensure that your cells are FYA/FYA and not FYA/FY. Remember, anti-Fya will often show "dosage".

Also remember that not all Fya+b= cells are homozygous for Fya. If the donor is of African-American heritage they may be Fya+Fy4+.

Yes, that is what I meant by FYA/FY. The phenotype Fy(a+b-) may actually be heterozygous expression of the Fy(a) antigen (and I know that I've used bad terminology - one cannot have heterozygosity of an antigen, only a gene!).
